Novel Molecular Recognition Mechanism by Chemical Sensors

研究の背景 Background

蛋白質やDNAに代表される生体高分子は複雑なシグナルネットワークを形成します。このネットワーク形成には特異的かつ制御可能な分子認識が重要となります。これまでその多くは分子形状の相補性による結合として理解されてきました。近年、化合物の形状ではなく電子状態を認識するケミカルセンサーが発見され、その特異な分子認識機構が創薬の分野からも注目されています。

Highly complicated signaling network consists of bio-macromolecules and is based on the highly specific molecular recognition. Molecular recognition by shape complementarity (Lock and Key theory of enzymes etc) has been widely studied. Recent studies showed that the chemical sensor protein recognizes electrophilic property of various small molecules and its novel molecular recognition mechanism has been attracting attention in the field of drug discovery.

研究の目標 Outcome

センサー蛋白質が求電子性を有する低分子化合物を認識する分子機構を明らかにするために、ケミカルセンサーと低分子化合物複合体の単結晶X線構造解析、熱力学的パラメータ変化の解析などを実施します。これらの解析結果と分子動力学シミュレーションを組合せて、ケミカルセンサーによる新規分子認識機構の分子レベルでの解明と創薬への応用を目指します。

To clarify the novel molecular mechanism of electrophilic small compounds by the chemical sensor, we study the crystallographic and thermodynamical analyses of the sensor and the small molecules complex. We also analyze the molecular dynamics simulation of the complex and unveil the novel molecular recognition mechanism of chemical sensor for various electrophilic small molecules.
