Structural and mechanistic studies of Ca2+ homeostasis in the endoplasmic reticulum

研究の背景 Background


The most important function of SERCA is to transport Ca2+ from the cytosol to the ER lumen. The Ca2+ transport cycle of SERCA consists of multiple reaction steps, during which the Ca2+ pump undergoes large conformational changes. Due to the limitation of crystallography, the known structure of SERCA1a in each reaction intermediate is fixed in a single conformation, and, in some cases, biased by crystal packing, which limits our understanding for molecular mechanisms of SERCA.

研究の目標 Outcome


We determined high-resolution structures of SERCA2b in various intermediate states by cryo-EM. By further investigating conformational dynamics of SERCA2b during the catalytic cycle, we aim to comprehensively understand the molecular mechanism of SERCA2b-mediated Ca2+ transport into the ER.
